The two females were aged in their nineties and died in residential aged care.
Premier Jeremy Rockliff expressed sympathy to the families.
"I extend my sincere condolences to the women's family, friends, and loved ones in this extremely difficult and sad time," he said.
More than 1 200 Tasmaians were diagnosed with the virus on Thursday, with a total of 8 609 active cases across the state.
Mr Rockliff said cases are trending downwards across the last seven days, compared to peaks seen earlier in April.
"We are continuing to see day to day fluctuations in case numbers, but overall, cases are trending downwards," Mr Rockliff said.
"Our hospitalisations remain among the lowest in the nation, and we expect to see the number of people in hospital for COVID decline over the next few weeks."
There were 1 265 new cases yesterday.
There are a total of 2 255 active cases in the North, 1 193 in the North West and 4 737 in the South.
"The number of people in hospital with COVID has also decreased to 41, with 12 of these being treated specifically for COVID, and just one currently being cared for in the ICU," Mr Rockliff said.
"The most important thing Tasmanians can do is to continue to roll up their sleeves and get your vaccination or your booster if it's due, to keep yourself and the community safe and continue to practice COVID safe behaviours."
