The new return service, which will take off from November 5, includes three flights on Monday, Friday and Sunday in each direction.
The company's announcement means Northern Tasmania will now have a direct connection to every mainland state capital following the launch of Virgin flights to Adelaide earlier this week.
Virgin Australia's group chief strategy and transformation officer Alistair Hartley said tickets are now on sale, with one-way flights starting from $149.
"We're continually looking at ways to pivot and expand our network schedule where free movement of travellers across borders permit, so we're incredibly proud that by working with Launceston and Perth Airports we've been able to launch direct services between the cities in time for the busy Christmas holiday period," he said.
Launceston Airport chief executive Shane O'Hare said the newest Adelaide route, commitment to Perth and an increase capacity to existing Brisbane flights will see 3000 new seats filled at the airport each week.
"This month's domestic growth coupled with the airport's increased airline capacity during the pandemic prove Launceston is a rising tourism destination especially for the long-weekend leisure market," he said.

"The new airline services will help revive our airport workforce as we progressively welcome back more staff.
"Western Australia has long been on our radar as an untapped market for the north of Tasmania, and we look forward to enabling frequent travel opportunities for people at both ends.
