Saturday's 41-point win over Burnie at West Park also saw the Two Blues collect just its third minor premiership ever, with the other two coming in 1932 and 1982 for a club that has been around since 1890.
Two Blue Jack Templeton, who managed two goals in the win over the Dockers, also locked in the club's first league leading goal kicker in its NTFL/NWFL history.
"It's awesome - we did the work and we deserve to be on top but the job isn't done," coach Maverick Weller said.
"We're really confident with our form lately that our game sticks up and we have some players coming from injury.
"We're in a really good spot and players have bought in. And it's a really special time because it's the first time for 40 odd years."
Penguin will host Devonport next weekend for a spot in the grand final, with Wynyard to welcome Burnie to its home ground for a cut-throat semi-final.
The Two Blues defeated the Dockers 13.21 (99) to 8.10 (58), with Luke Russell Dusten O'Neil and Dylan Bramich playing key roles.
The Dockers were best served by Seth Campbell, Joshua Wolfe, Harry Walters, with Isaac Plant kicking three goals in what was Rudy Barrett's 200th game for the club.
Wynyard got a much needed win when it made the trip to Latrobe.
After dropping four of their past five games, the Cats defeated the Demons 15.9 (99) to 10.10 (70) after a slow start.
Kallum Kubicki, Matthew Bugg and Jonty McIvor were Wynyard's best, with Zac Roberts, Brayden Scott and Shaun McCrossen strong for the Demons.
Devonport had an easy 121-point win over wooden spooner Ulverstone, 20.13 (133) to 1.6 (12).
