The airline's announcement came after it was confirmed Tasmania's border would reopen to New South Wales on November 6.
From December 4, Qantas will operate three return services each week, before increasing it to four return services the following week until February 1. This will bring about 900 passengers each week to the city.
It's the first time since 2004 the airline has offered the route and it will look to add more flights beyond the eight-week period if there is demand.
Qantas chief executive officer Andrew David said the company was delighted to add the direct flights to its roster for the first time in 16 years.
"We know there is huge pent-up demand for domestic travel with Australians looking for new destinations to explore," he said.
"These new services to Launceston will help more people discover Tasmania's beautiful north and give locals more options to get to Sydney.
"We'll keep a close eye on demand and if it proves successful we will look to extend the service."
IN OTHER NEWS:
Regional Tourism Assistant Minister Jonno Duniam said it was fantastic news for the state's tourism industry.
"This summer is going to be critical in our state's tourism recovery, and this new route opens Tasmania's north to more visitors just days before Christmas," he said.
"I can't wait for Sydneysiders to see for themselves the best beaches, wineries and wilderness our country has to offer, right here in Tasmania."
Qantas is offering special fares starting from $149 one-way until Friday.
The company will also restart flights between Hobart and Sydney, with 11 return services per week from November 6.
