A patron entering the Victoria Parade business reported a fire in room 22 to staff, about 7.45pm on Friday.
Firefighters were called and contained the blaze to the one room, which is next to other rooms, but separate to the main hotel building.
No one was injured, but there was extensive damage to the unit.
Fire investigators determined that the fire was deliberate.
A Tasmania Fire Service spokesman said damage was estimated at $50,000.
Detective Senior Sergeant David Chapman, of Western Criminal Investigation Branch, said anyone with information about the suspicious fire should call detectives on 6478 4003 or Crime Stoppers on 1800 333 000.
