The show will be performed four times in three days by Launceston Church Grammar School's grade 6 pupils with the first slot scheduled for 1pm on Tuesday.
Long-time school music teacher Mark Saunders will direct the performance.
He said the junior version of the 1988 Roald Dahl classic book is a great production and a suitable level of difficulty for children of this age.
"Viewers should expect 75-minutes of absolute fun, singing and dancing," he said.
"It's just a great story of children who try their best to misbehave in order to get back at their teachers."
In total, 44 pupils, who underwent auditioning for each of their roles, will be utilised for the performance.
The role of Matilda's beloved teacher Miss Honey will be performed by Alice Gannon, 12, who said she is excited to finally perform.
"The experience has been really great so far, I'm a little nervous but Miss Honey is such a fun character to play," she said.
"I've been trying to practice every night, but if I can't, I make sure to at least do it every second night."
According to teacher Helen Wilson it has been a running tradition for grade 6 students to put on a theatre production.

"The whole process is about everyone having a role so that they're able to practice working together while learning the craft of theatre as well," she said.
"The kids are loving it, I think the professionalism of lights and audio has really impressed them and they've taken it all very seriously as a result."
She said the production has been a collaborative effort between the school community and commended parents for their efforts in helping out wherever they can.
"They've helped with costumes, hair and of course with helping the kids practice, I can't thank them enough," she said.
The event's audience will consist of friends, relatives and fellow classmates of the pupils involved. Anyone else that would like to attend is urged to contact the school.
The production will finish with its final performance on Thursday, September 23 at 7pm in the school hall.
