The fair moved online last year due to COVID-19 restrictions, but will be back as a physical event this year.
Located at Deloraine, the annual event showcases a mix of artists and craftspeople, and provides them with a platform to sell or demonstrate their work.
Craft Fair director Rob van der Elst said with the fair celebrating 40-years, this year's event was a big deal.
"We really want to push that and to celebrate we have an exhibition on with four artists who were involved in the early years of the craft fairs and have become well-known," he said.
"I think it's become a bit of an iconic event over the years and we have a lot of people who are regulars, both stallholders and patrons."
The fair will consist of about 200 stallholders, with around 40 of those from the mainland.
Mr van der Elst said the stallholder program may not be released until closer to the date due to border concerns for some of the mainland participants.
The event has a detailed safety plan and includes mask-wearing, after Premier Peter Gutwein announced they would be mandatory at events with more than 1000 people.
Mr van der Elst said attendees of the fair were encouraged to bring their own masks as supplies at the event would be limited.
"We are very proud [the fair is going ahead]. We talked about it earlier on in the year and there was a bit of doubt here and there, but we have to keep going and if something happens we just have to carry it," he said.
Tickets for the event are available online and through the visitor's centre at Deloraine, and buses will run between venues.
Mr van der Elst hoped the community would support those who did not have many opportunities to sell their work elsewhere by attending the event.
The 2021 Tasmanian Craft Fair will be held from October 29-November 1. For more information visit tascraftfair.com.au.
