He will not be present for the final day of Parliament this week, and deputy premier Jeremy Rockliff will take on his duties for Question Time and other matters before the House of Assembly.
Mr Gutwein said he would continue to carry out his other duties as Premier while at home.
He has taken a rapid antigen test which came back negative, but under Public Health guidelines he must isolate for seven days.
It comes at the end of a challenging morning for the Premier during which his government faced sustained questioning from Labor over a lack of apology from Attorney-General Elise Archer and senior cabinet minister Michael Ferguson to a victim-survivor of child sexual abuse.
Mr Gutwein accused Labor of "making up" allegations that Ms Archer and Mr Ferguson had groaned at a question asked on behalf of the victim-survivor, Tiffany Skeggs.
Speaker Mark Shelton then gagged debate twice.
