About 70 participants will pedal more than 400 kilometres for the cause, which this year aims to raise $50,000 for Down Syndrome Tasmania and Muscular Dystrophy Tasmania.
Now in its 13th year, Northern Senior Sergeant Nick Clark said the ride was about raising awareness and supporting those in need.
"Over the years we have supported numerous charities," he said.
"It's all about making people in the community aware of these conditions and fundraising to make a difference to those impacted."

This year's participants include about 35 Tasmania Police members, along with representatives from businesses and communities from all over the state.
Since its inception in 2007, the event has raised more than $500,000 for Tasmanian causes. With this year's ride to take place from November 28 to December 1, Senior Sergeant Clark encouraged everyone to keep a look out for the team.
"The welcome we get from each community as we pass though makes it all worthwhile," he said.
"The satisfaction is knowing we are doing our bit to help. This is the third time we have supported Down Syndrome Tasmania.
"The interaction we have with the kids, who often meet us on the ride. The looks on their faces is always amazing."
