A rider and the pillion passenger were involved in the crash at Berriedale following the Motorcycle Riders' Association of Tasmania 2013 Toy Run.
The motorcyclists were in the north-bound lane of the Brooker Highway when the crash occurred at 5pm. Police did not release their names or further details last night.
Motorcycle Riders' Association of Tasmania South president Peter Dixon expressed his condolences.
``My deepest feelings go out to the families involved,'' he said.
It is the second fatality in as many years related to the toy run.
North-West woman Kim Dougan died at Conara on her way to the Southern event last year.
More than 6000 bikes took part in the event, which sees riders meet at the Derwent Entertainment Centre before distributing toys through Hobart.
Donations, which included food, clothing and toys, were given to the Salvation Army.
