Tasmania Police and fire crews were called to Seaz Takeaway at Alanvale Road, Newnham around 10.20am on Sunday to find the business and an attached weatherboard house well alight.
Thick smoke could be seen from across the river and forced several nearby residents outside after being "smoked out" of their homes.
Six fire crews worked for over two hours to get the blaze under control, after fuel load and construction difficulties hindered their efforts.
Several firefighters were treated by paramedics at the scene, but no serious injuries were reported.
One neighbour said she was saddened to see the local institution go up in flames.
"I'm just devastated, they make the best fish in chips in town," she said.
"Their fish is just amazing, they have the best portions."
A spokesperson for the Tasmania Fire Service said the fire had been investigated and deemed accidental.
"The fire started in the kitchen of the adjoining house caused by an unattended pot on the stove," they said.
"The resident had attempted to extinguish the fire but was unsuccessful.
"Everyone was safely evacuated from the house and the shop."
The spokesperson said the fire was a reminder to remain vigilant while cooking, especially when cooking with oil.
The TFS also thanked a good Samaritan who had been in the shop at the time and helped people to evacuate the building swiftly.
"A customer in the shop at the time of the fire was instrumental in evacuating people from the house prior to the brigades arrival."
A neighbour said he hoped the business would reopen in another location.
"It's just so devastating to see, someone's whole livelihood being destroyed like that," he said. Damage to the property has been estimated at approximately $500,000.
