All cases have links to Newcastle, the location of a superspreader event at a nightclub, Premier Peter Gutwein said.
Tasmania threw open its borders to all fully vaccinated travellers on Wednesday, with one case recorded on an early-morning Qantas flight from Sydney.
Mr Gutwein said a further two cases had been recorded in the 24 hours to 10am on Friday.
One of the new cases was on the same flight, while the other travelled to Tasmania from Melbourne on Wednesday afternoon.
State Public Health Director Mark Veitch said the two new cases have had very little interaction with the community and went to get tested soon after being told they had been at an exposure site.
They tested negative before travelling to Tasmania.
All recent arrivals from Newcastle are being contacted by public health and being asked to get a test.
Tasmania requires people arriving from high-risk areas to return a negative test in the 72 hours before they travel.
Mr Gutwein said the state government was considering a mask mandate at indoor venues and would have more to say in coming days.
Tasmania had no active cases when it reopened.
