The siblings established Love Your Sister with the aim of "vanquishing all cancers, now!" in Connie's words.
Mr Johnson said he was excited about this weekend's three Launceston fundraising events.
"To be doing her work on her anniversary is only fitting, so I'll be celebrating by doing exactly what she told me to do - raising funds for cancer research," he said.
IN OTHER NEWS:
Love Your Sister has established a million-strong community who have been raising money and awareness around the country.
"The community response to this fundraising tour surprises me every day. It's been a combination of two things, learning how to do my job well and that there's way too many families similarly affected by cancer," he said.
"I'm overwhelmed by the love I'm shown on the day."
Mr Johnson and this community have raised $10 million at more than 800 community events, with the Relbia event organised as an extra special Toast to Connie, Josef Chromy Wine Club and marketing manager Amy Russell said.
"It will be a fairly emotional day for Samuel," Ms Russell said.
"He has been doing little pop-up fundraisers around the country. We were more than happy to host it here and we'll have a toast to Connie so everyone can join in," she said.
Josef Chromy Wines is selling $5 raffle tickets to win a private tasting with Sam for up to four people, with all proceeds going to Love Your Sister.
The business will also donate $2 from every glass of Pepik Sparkling Rose sold and 20 per cent of the value of wine orders placed during the fundraiser.
The event will include a presentation about Love Your Sister, special toast to Connie and the private wine tasting for the raffle winners.
Love Your Sister wants to raise $1 million a month by December 2020.
"I need 100,000 Aussies in at $10 a month. I've built a Cancer Bank and I'm keen for investors!"
