NSW were dismissed right on tea at the Gabba for 298 for a handy 263-run lead, on what proved to be a tough wicket to score runs on across the opening two days.
The Blues' innings was built around half centuries from Moises Henriques (78) and Daniel Hughes (68), with the latter grinding his way through 218-balls in energy sapping conditions.
Henriques scored 11 boundaries in his 116-ball knock as the pair put on a match-high partnership of 119 for the third wicket.
Jack Edwards added an invaluable 40 at No.7, while Trent Copeland contributing a run-a-ball 22.
Michael Neser (3-73) was the pick of the bowlers, picking up Hughes in the final over of the first session and knocking over the consistent Peter Nevill (9).
Batting conditions on a green-tinged Gabba wicket have improved after the opening two days produced 22 wickets.
Fourth-placed Queensland can ill-afford to lose against the second-placed NSW with only two rounds remaining.
