Coach Ken Morton will be wary of the Strikers but is confident his side can win the title in front of its home crowd.
"This is a massive game for us on Saturday, we need to win to be crowned league champions," Morton said.
"Winning the premiership on your home ground would be fantastic for the whole club, players and supporters alike."
Goalkeeper Kane Pierce returns from suspension while Ben Hamlett should be fit and Andy Brennan will play after an easy week on the training track.
"It will be important for us to be very compact defensively and to open up the spaces with quality passing and movement when in possession of the ball."
Devonport coach Nick Owen was pleased with last week's win but knows South will be a different opponent entirely.
"Last week's performance was good, but beating a team lower on the ladder at home is a lot different than taking on the top of the table team only needing a point to defend their title," Owen said.
"South have been dominant again this year and their squad only seems to grow stronger. Our focus for the remainder of the year is to play attacking football and this weekend will be no different."
The two Launceston-based sides have home games, Northern Rangers taking on Kingborough Lions and Launceston City hosting sister club Hobart Zebras.
A win for Rangers would seal its spot in the finals series.
"This week is certainly about getting the points, but it is also about continuing to build a better team performance each and every week, as this is what we are aiming for," coach Tim Lunnon said.
"We are asking the players to work harder for longer, as this is what competing is about, and if we can do this combined with our team play then we should continue to improve our performances."
City's previous match against Zebras ended in a humiliating 12-0 defeat, but coach Kurt Reynolds said his side had nothing to lose.
"[Zebras] are a good football side and the match will be a challenge, [but] this is another opportunity for us to move off the bottom of the ladder through a positive result," Reynolds said.
However, the home side has injury concerns to Toby Omenihu, Andy Compagne and Aaron Mattarozzi.
The round kicked off last night with Glenorchy Knights taking on Olympia at KGV.
Knights are playing for pride and need two wins from their last three games to equal their points from last year.
