Porte crossed the line in three hours, 42 minutes and 20 seconds, some ten seconds ahead of Italian Diego Ulissi and fellow Australian Simon Gerrans with Cadel Evans sixth, 14 seconds behind.
Porte broke away in the last 2km of the Old Willunga climb in a masterful ride.
Gerrans has regained the Tour lead from Evans by one second going into tomorrow's crucial final stage.
Porte is fourth in the overall general classification 10 seconds behind Gerrans.
He is third in the King of the Mountain category with fellow Tasmanian Will Clarke just behind him in fourth and ninth in the battle for the sprint jersey.
If Gerrns can hold his lead in the final stage, he will become the first three-time winner in Tour history.
