Emergency services were called to the crash at Travellers Rest by a member of the public about 4.40am on Thursday.
Tasmania Police Sergeant Gavin Chugg said it appears the car has left the side of the road and hit a cement curb.
Police are searching for the driver of the Mazda hatchback.
“Given the serious nature of the crash, it is possible the driver could have serious injuries,” Sergeant Chugg said.
One lane of the Highway is open, but traffic is moving very slowly.
Anyone travelling in the area can expect delays until the scene is cleared.
Investigations into the cause of the crash are ongoing.
Information about the crash should be reported to police on 131 444, or anonymously to Crime Stoppers on 1800 333 000.
