The Acting Attorney-General announced his decision on Saturday alongside Premier Will Hodgman.
He will serve for the remainder of his current term as Member for Denison, but will not continue his ministerial duties.
The father-of-three said the decision was a difficult one, but that it was the right time for him to move on.
Mr Hodgman said the decision came as a surprise to him, but that he understood his reasons.
“Politics can be extremely demanding and stressful on family life, and I am sure all Tasmanians will understand Matt’s decision,” he said.
While in government, Mr Groom has served as a Liberal parliamentary member for nearly eight years, after being first elected in 2010.
He has held a number of portfolios, including State Growth, Energy, Environment, Parks and Heritage, as well as recently serving as Acting Attorney-General and Acting Justice Minister.
Mr Hodgman said a reshuffle of ministers would be announced “in due course”.
