Van Diemens Street Rod Club president Grant Sutton said more than 200 vehicles would be on display, including muscle cars, classic cars and hot rods.
Mr Sutton has been involved in the club for more than 30 years.
"I just like all forms of motor sport, and this is a way for people to display their passion - it's a growing passion," he said.
"This year, we've opened up the paddock next to Aurora Stadium again, so we'll have the best of both worlds."
Launceston resident Bill O'Connor will display his 1955 Ford Thunderbird for the first time at the show.
He said he bought the car in 2013 and had been restoring it ever since.
Mr O'Connor and his Thunderbird will travel to Melbourne for a hot rod show after the USA Day All Makes Car Show.
There will be food vendors and a DJ at the event, which will run from 10am to 3pm. Money raised from the car show will support the Cancer Council.
Entry costs $10 for adults, with children under 14 admitted free.
