The win made it 10 in a row for the Tornadoes, who will be on a tight turn-around to play against the Geelong Supercats in Geelong on Sunday.
The Tornadoes scored 80 to the Chargers' 52, with eight individual scorers putting points on the board.
Tornadoes coach Reece Potter said it had been a "massive" win.
"I think as a team our ability to play together and execute together and defend for each other really showed in this game," he said.
"It was a really nice team win, and a 28-point win against Hobart in Hobart you would take any day of the week.
"Every game I'm quietly confident that we can, but I also know that if we don't turn up or we're not ready to go we can easily get beaten by any team we play [because] it's such a strong league."
Potter said star performers included Mikaela Ruef, Lauren Mansfield and Mariah Payne.
"I think overall it was a team win, eight scorers throughout our team, multiple contributors on the defence advance, and like I said our depth against Hobart showed," he said.
"We had more players coming in and contributing than they did, and that was a really key part of us winning the game tonight."
