Clarence won the inaugural statewide competition and is favoured to win consecutive premierships, but Devonport will be eager to wrest the title from the Roos.
AFL Tasmania chairman Dominic Baker said that the competition had been healthy and vibrant this year.
"It's obvious to me that the top two teams of the competition are fully deserving of the opportunity they have earned to win the 2010 premiership," he said.
Grand final day starts with the reserves match between Clarence and North Hobart at 9.05am, followed by the under-18 game between North Launceston and Clarence at 11.15am.
Clarence and Devonport face-off in the big game from 2.05pm.
The remaining top 10 players in the Tassie Medal have also been announced, with a host of players still in the running.
Six North and North-West players and four Southern players - including last year's winner, Kurt Heazlewood - are still a chance to win the award in which six of the league's 10 clubs are represented.
Clarence coach Brett Geappen is still in contention, despite his retirement midway through the season due to injury.
Tasmanian State League general manager Shaun Young said that it was a glowing testament to the competition that there was an even spread of northern and Southern players still in contention for the medal.
