De Winter has been the Tigers' interim head coach this summer following the resignation of Jeff Vaughan to take up an opportunity with the national side, with this week's Sheffield Shield clash with Queensland set to be De Winter's last in charge.
"This will be my last game,'' he said.
"I've had a lot of thought bubbles in the last 10 or 12 days and I just feel like this is a good time to move away and let someone else come in and take control of what is a really good stable group now.
"We don't think we need to add much more to the squad, but I will stay around the group and support where I can, but is an ideal time to hand it over.
"What was three months has turned into nine or 10 months and it has turned into the greatest ride I've had on my cricket journey, and I'm happy to tick that box and its an itch that has been scratched."
De Winter, who himself has worked with the national team, said he still felt he had a lot to give from a coaching perspective, but not as a head coach, and was looking forward to supporting whoever took up the role from a Tigers and a Hurricanes point of view.
Nathan Ellis and Ben McDermott will be unavailable as they are part of Australia's limited overs squad for the tour of Pakistan, while RIley Meredith and Matthew Wade have headed over to the IPL.
Veteran paceman Peter Siddle will be rested and will join the Tigers' coaching staff for the match against Queensland.
The clash with Queensland at Blundstone Arena will start on Wednesday.
