About 9am on Saturday morning, emergency services were advised of the incident.
A grey Mitsubishi Triton turning from Elderslie Road into Rodbourne Road collided with a white Toyota Corolla travelling towards Brighton.
As a result of the impact, the Corolla rolled onto its side and struck a vehicle waiting to turn from Rodbourne Road on to Elderslie Road.
The driver of the Corolla was declared dead at the scene, and the four occupants of the other vehicles were treated for minor injuries.
Elderslie Road was closed for eight hours while investigators assessed the scene.
All drivers involved were subject to mandatory drug and alcohol testing.
The cause of the crash is under investigation, but Tasmania Police said early indications suggested speed and inattention were contributing factors.
Police are asking anyone who saw the Toyota Corolla or the Mitsubishi Triton travelling in the area prior to the crash to contact Crash Investigation Services on 131 444.
