The crash occurred yesterday about 2.10pm when a truck travelling towards Hobart collided with a small sedan heading north.
The two male and female occupants of the sedan were killed instantly.
Tasmania Police are continuing to investigate the double-fatality today, which saw the state's death toll rise to 20, two higher than the same time last year.
A police spokesperson said they were continuing the process of formally identifying the victims and notifying their next of kin.
