In a statement, Tasmanian Police said that a man and a woman had died in the two-vehicle crash.
"The deceased people were travelling together in the same vehicle," the statement reads.
"The Bass Highway remains closed while crash investigators examine the scene. Traffic diversions are in place."
Emergency services were called to the scene just after 11am.
The Westpac rescue helicopter had earlier been dispatched to the scene.
Motorists are asked to avoid the area, and divert at the Bass Highway on Meander Valley Road or the Bowerbank Link.
Police will provide further information about the crash later this afternoon.
