Mr Duigan defeated Labor's Geoff Lyons and independent candidates Will Smith, Rob Soward and Vivienne Gale for the seat.
Speaking from his motel room in Melbourne where he is filming for TV show Hook Line and Sinker, Mr Duigan described the day waiting for results as a rollercoaster.
"It's completely overwhelming, it's been a really long and at times anxious wait," he said.
"We always knew the preference split up would be today, and it's been a really tense day with a range of emotional ups and downs, it's been a rollercoaster really.
"I feel somewhat relieved, but a bit overwhelmed."
IN OTHER NEWS:
Mr Duigan said it was humbling to be given the responsibility by Windermere voters, to be so "graciously and roundly accepted".
"I don't have any legislative experience but people have seen something in me and have voted in good support of me to give me a crack at this, and I'm really conscious of that responsibility," he said.
"I just want to do a really good job for the people and the voters of Windermere, they are the ones who tasked me with this and I want to do them proud."
Celebrations will be subdued for Mr Duigan tonight.
"I've had the opportunity to speak with my wife and my children and some close friends and supporters to thank them for all their support and their generosity along the way," he said
"I haven't really allowed myself to go any further [thinking and planning] than this day, allowed myself to think what this would look like if I was fortunate enough to win, now I'll have to really give those things some consideration."
Liberal party state director Stuart Smith said Mr Duigan had been an outstanding candidate.
"The Tasmanian Liberals would like to thank the thousands of Liberal supporters who have voted for us in Windermere, and express our gratitude to the many volunteers, supporters and members for their help in winning the Party's fourth seat in the Legislative Council," he said.
"Congratulations to Geoff Lyons and Will Smith for their determined local campaigns in Windermere.
"We wish them well in the next endeavours."
