Shane Navin, 68, of NSW crashed 35.4 kilometres into the fifth stage about 10am on Friday and died at the scene.
The 1979 Mazda RX-7 reportedly rolled while navigated a right-hand bend. Co-driver Glenn Evans escaped without injury.
Targa Australia CEO Mark Perry said the death of a driver in the Tasmanian race was "very sad" for all involved.
IN OTHER NEWS:
"Shane was a much loved and admired member of our Targa family," he said.
"We send our heartfelt condolences to Shane's family and friends on their loss."
Tasmania Police Inspector Gary Williams said investigations into the cause of the crash were continuing with a report to be prepared for the coroner.
"I am aware that the vehicle did roll and it rolled into a culvert, but the cause of the accident will be a matter for the coroner and is currently being investigated," he said.
"I understand it was a right-hand bend and something has occurred whilst transiting that right hand bend which has led to the crash."
Inspector Williams said the crash served as a reminder that driving was "risky business" regardless of a driver's experience.
"Anyone who has driven those roads know they are particularly challenging roads, they are very windy, quite elevated and they can be challenging roads," he said.
In a separate crash on Friday, a truck driver became trapped and sustained serious injuries to their legs after crashing with another truck on the Bass Highway at Sisters Hills about 9.30am.
Inspector Williams said the driver was flown to North West Regional Hospital and would require surgery.
"Early indications appear that one of the trailers attached to the prime movers may have swung out into the path of the truck coming the other way," he said.
"You have got two very heavy vehicles travelling at highway speeds and if they impact each other head on that is going to cause significant damage."
