Campania and Bothwell kick off the round on Friday night at 6.30pm with the home side looking to bounce back from last week's opening round loss with the inclusion of 2019 Association Best and Fairest winner Matthew Triffett and Kyle Smith's vital midfield drive.
Brodie Howard battled hard in the conditions last week and should find plenty of the ball again, while Alex Langridge and Dean Wall provided invaluable experience for the young side.
Bothwell will be coached by 2015 association and five-time club Best and Fairest winner Simon Bryant and will lead a team with plenty of potential to be a top side.
The club has recruited Justin Brown from Swansea, Kyle Clark joins as assistant coach and Hayden Graham crosses from Oatlands.
Tom Steele returns from Lauderdale and will be invaluable across half back while premiership forward Kris Cashion also returns and will be a focal point in front of goals.
Campbell Town plays host to Mt Pleasant on Saturday night at 6.30pm.
The Robins had a good opening round performance, proving they are not to be taken lightly and will improve as the season progresses.
Matt Carlile had an immediate impact through the midfield in his return to the club and was ably supported by Josh Townsend.
Jamin Douce will look to lock down the Mounties' forwards while Angus Chugg should generate plenty of tap advantages.
With inclusions Kayden Douce, David Watkins and Dylan Westlake, the side should make a fast start after last week's opening quarter performance.
Mt Pleasant had a dominant first round win and will look to make it two from two with Mitchell Byers a standout from last week in tricky conditions.
Ross Hugo and Nicholas Davis were impressive in their first games for the club and have added to the depth on the field. Injuries will result in some changes with Brad Duggan, Jamie Duggan and William Iles expected to join the line-up this week.
