All four of the state's planned relays will go ahead with government-approved COVID-19 plans, with numbers to be capped at 2000 people.
This includes participants, vendors, staff and volunteers, for each event in Launceston, the North-West Coast, Derwent Valley and Hobart.
Relay For Life state manager Alicia Van Ek said after the event was cancelled in 2020, organisers had since gone above and beyond to ensure the major fundraiser went ahead this year.

"Everyone has become really accustomed to COVID and all the restrictions that it brings," she said.
"We are legally obliged to have a COVID safety plan, but we are going above and beyond when it comes to safety.
"We have very vulnerable people with compromised immune systems that are attending this event, so we aren't taking any risks."
Ms Van Ek said a ticketing and registration process would be implemented to ensure the contact details of each patron is registered, along with a separate entry and exit point to count the number of patron's onsite at any one given time.
Increased frequency and standard of cleaning as well as additional hand sanitiser and soap will also be made available onsite.
Relay events will begin in late February in the Derwent Valley and finish on the North-West Coast at Penguin in late March.
Cancer Council Tasmania chief executive Penny Egan said staff, the committee and volunteers were all being provided COVID-19 training to ensure events were managed safely.
"The reality is we will be living with COVID-19 for some time, even as the vaccine is rolled out Australia-wide. We want our relayers to know they will be safe throughout our major events," she said.
"Relay For Life is not just an event - it's an experience that provides lasting memories and people who participate know that the funds they raise stay in Tasmania.
"For every person who is walking with their mother or sister or father who has survived a cancer diagnosis, there is a group alongside them who are walking in remembrance of someone who has lost their life to this disease."
