The 17-year-old dominated the women's event, carding the joint best score of 70 in the third and fourth rounds to win by a huge 10 shots from Ulverstone's Sarah Johnstone.
A previous winner of the Tasmanian Junior Masters and the Judy Elphinstone 54-hole tournament in 2019, Wilson's performance left her at even par for the 72 holes and was the biggest victory for the Rosny College student.
"I was a little nervous going into today, but not very," she said. "This tournament, it's all just come together. I didn't hit it as well as yesterday, but I was steady."
Wilson hopes to pick up a college scholarship in the United States after completing her amateur career.
Meanwhile Queensland's Louis Dobbelaar prolonged his winning habit by taking out the men's event.
The 19-year-old has previously won two Queensland Amateurs, the New Zealand Amateur and the Port Phillip Amateur in Melbourne.
Dobbelaar carded a one-under par 71 on Monday to go with earlier rounds of 67-65-70 to win by eight shots from last year's winner, another Queenslander Will Florimo.
Launceston's Greg Longmore was the best-placed Tasmanian finishing fifth on +1.
In the Tasmanian Senior Masters, legendary Lindy Goggin closed out the women's section with the best round of the week, a 78, to win by a massive 13 shots from Cheryl Richards, of Mowbray.
In the men's masters, Robbie Berne (Prospect Vale) won by seven shots over the 54-hole event.
