New Wallabies coach Dave Rennie has unveiled a mix of youth and experience through the side, handing three Queenslanders their first start.
Harry Wilson, aged just 20, is at No.6 while winger Filipo Daugunu and outside centre Hunter Paisami are also in the run-on side.
While Paisami got the nod with Jordan Petaia still returning from a hip injury, Rennie said the other two were irresistible selections.
"Harry has been a real stand-out - he's a big ball carrier, aggressive defensively and we were keen to have him and Pete Samu on the park because they provide a point of difference," Rennie said on Friday.
"Hunter, with Jordie being unavailable, is an aggressive ball carrier and really destructive defensively and we think that's going to be important.
"Filipo, I thought he was just outstanding for the Reds - he's a real handful and can beat people in a phone box and is really aggressive defensively.
"They were pretty straight forward decisions to be honest."
Rennie said that while Wilson spent the season at No.8, he thought the speed of Samu could be a "weapon" off the back of scrums.
James O'Connor will start at five-eighth with Lolesio eased into Test action, while another veteran Matt Toomua will partner 22-year-old Paisami in the centres.
Nic White is the starting halfback while Waratahs No.9 Jake Gordon is in line for his second cap among the reserves, with Tom Banks at fullback with Dane Haylett-Petty injured.
"James got better and better at 10 and I've been really impressed with him as he understands the game inside out and has been demanding of others," Rennie said.
"With Hunter and Filipo on the field we thought it was important we added a bit of experience around them."
After two years in the Test wilderness, Matt Philip will add to his three caps when he takes his place in the second row at Sky Stadium next to Lukhan Salakaia-Loto, while Pete Samu is at No.8.
Rounding out the back row is captain Michael Hooper, playing his 100th Test.
The front row sees 96-Test veteran James Slipper and Taniela Tupou alongside hooker Folau Fainga'a.
