Olympic's Taylor Neilson and City's Sam Ridgard have both missed several weeks due to persistent leg injuries.
But only Ridgard has a chance to return for the 2.15pm clash at Windsor Park.
The English striker has not featured since hurting his ankle midway through the last round of the Lakoseljac Cup but hopes to be back this week.
"I'm not 100 per cent over it but I've trained on it and should be able to play," he said.
"For us it's a must-win game. It's a derby and after everything that happened in the last game with Rob (Gerrard) getting suspended for an extra game, there's a lot riding on it."
A strained adductor expected to keep Neilson out for a week has stretched that out to three.
"It's really frustrating not being able to contribute," he said.
"But Max Reissig has done well. He's playing my position and doing it properly, tracking back and getting forward a lot more than I do.
"This is the match you mark on the calendar - it's the most important for both our sides."
After a red card on Saturday, the versatile Matty Oh is suspended for City while Spanish playmaker Nil Sanz is back for Olympic.
The sides will play for a cup provided by Guardian Pharmacy Prospect which also supports Cavaliers who will feature in the State Netball League grand final being played simultaneously at the Silverdome.
