Perennial title contenders South Hobart and Devonport visit Launceston United and City respectively while Riverside and Northern Rangers both head to the capital.
Launceston United v South Hobart
United are proud to be hosting Ken Morton's big guns but coach Glenn Reading would rather have waited a few weeks.
"Originally, the game was going to be about four weeks into the season but now it's first up and I think we're going to be a bit underdone," he said.
"We've only been back training for three weeks whereas they seem to be playing a couple of games a week.
"We're going to be focusing on the Northern Championship but this will be a yardstick for us and we'll go in with a positive attitude."
Will Compagne has made a big impact since arriving from Launceston City and should play wide midfield supplying young striker Christian Byard, who scored nine goals for Burnie last season.
As a tribute to Liverpool's drought-breaking Premier League title, Reading plans to play captain Nathan Butler as a sweeper in the mould of Alan Hansen, the last Red to lift a top-flight title.
Glenorchy v Northern Rangers
Northern Rangers will be introducing some South American flair to Glenorchy in their fixture.
Hailing from Concon, on the Chilean coast west of the capital Santiago, Fernando Munoz is coaching last season's Northern Championship-winning side while living in Tasmania.
While his English may be a work in progress, his grasp of the coronavirus legacy is spot on.
"It is very complicated season," Munoz said.
"Our players have less training and this first game of season but we go try for win.
"We have good players and had good results in all teams - men, women, under-18s - and try and get back for future to NPL. We wish we have good result and pass into next round."
Munoz confirmed keeper Zac Chugg will be his captain but added: "For this season our key player is the team."
University v Riverside
Riverside coach Alex Gaetani is navigating some choppy selection waters with commitments either side of the River Derwent.
The club's glut of blossoming youth players makes them an attractive proposition for the under-20 cup in which they will play at Clarence.
But Gaetani insists the Lakoseljac Cup fixture at University remains the priority.
"We have an extremely strong under-20 side and could be beating most teams but four or five of those are NPL regulars now," he said.
"I think we can manage both cups but we will always prioritise our best 11."
The likes of Jarrod Hill, Liam Gilmore, Jasper King, Aaron Kidmas, Will Humphrey, Fletcher Fulton, Mitch Lockhart and Matt Spanos could all still play under-20s.
"They won't be sitting on our NPL bench," Gaetani added. "If an under-20 is not starting NPL, they will be at Clarence."
Buoyed by pushing champions Devonport to the dying minutes in Saturday's 1-0 loss at Valley Road, Olympic welcome Mexican winger Jhostan Padron into the frame with returning club captain Taylor Neilson hoping to be fit for round one.
"University are one of the strongest Southern Championship sides and drew with Kingborough at the weekend," Gaetani said.
"They are aggressive in their pressing and will know that NPL sides like to play out from the back so it's going to be tough.
"We are looking forward to the trip down and fancy ourselves but it's a danger game. We want to get through so are playing our best 11."
Launceston City v Devonport
As Riverside focus on youth, their cross-town state league rivals Launceston City are building on experienced foundations.
Coach Roger Hardwicke said Lithuanian import Gediminas Krusa joins the likes of Sam Ridgard, Lindsay Millington and Rob Gerrard to bring maturity into his side.
"He can play multiple positions, is very composed on the ball and brings a bit of quality and technique," Hardwicke said. "He does not get flustered and has a great engine to get up and back and if he played for Devonport he knows what the league is all about and it's great to have him in the group."
City face league champions Devonport in the Lakoseljac Cup and Glenorchy in the under-20 competitiion, separated by four hours and 200 kilometres.
"We are confident in our ability, respect who Devonport are and what they've achieved over the last three or four seasons but we're not there to make up the numbers.
"We showed against Olympia on Saturday that we can make it difficult for any side in this competition and potentially come away with the win."
Statewide Cup
Prospect will also host a tasty Northern derby in the women's statewide cup as City's re-formed side welcome Ian Loft's powerful Northern Rangers outfit.
Barry Baker's team will be keen to make a statement after the club withdrew from both regional and statewide competitions in 2019.
Reigning Northern league champions Riverside and Metro will both play their first games in the competition at Windsor Park.
