The men, aged 51 and 21, were arrested on Tuesday morning at a property at Derwent Park.
The arrests were made in relation to the seizure of more than 700 grams of methylamphetamine by Tasmania Police from the Spirit of Tasmania in late December, 2019.
Both men have been charged with trafficking in a controlled substance.
They have been bailed to appear in the Hobart Magistrates Court on August 10.

The operation, targeting Bandidos members and associates in the greater Hobart area, was carried out in collaboration with the Australian Federal Police National Anti-Gang Squad.
The arrests follow on from the large-scale police operation conducted at Dover last Wednesday.
Five people have now been charged in connection with the investigation.
"This operation has significantly disrupted the criminal activities of those linked to OMCGs in Tasmania," Detective Acting Inspector Nikala Parsons, of the Serious Organised Crime Division said.
"We are continuing to work closely with our partners at the AFP NAGS to target criminal gang activities."
