The two-time championship winner confirmed on Sunday he'll race in Wednesday's fourth round of the simulated racing series.
He'll be one of three wildcards competing in the event along with American pair Joey Logano and Alexander Rossi.
"The Eseries has gained a bit of interest in its first three weeks and it will be good to be a part of it, racing against the regular Supercars guys and Joey," Ambrose told Speedcafe.com.
"I have done a bit of iRacing in the past, but I might have to do a bit of work in the next few days to make sure I am up to speed."
Ambrose will race in a Pirtek-sponsored Ford Mustang, similar to the Tasmanian's championship-winning Falcons from 2003 and 2004 before he went on to have a successful 10-year racing career in the United States.
NASCAR racer Logano will drive a third entry for DJR Team Penske alongside Scott McLaughlin and Fabian Coulthard while Indianapolis 500 champion Rossi is joining Chaz Mostert and Bryce Fullwood in Walkinshaw Andretti United Holdens for the event.
