Devonport and South Hobart were also finalists in three of the last four tournaments and will be facing away ties at Launceston City and United respectively.
City's newly-appointed coach Roger Hardwicke welcomed the visit of the reigning NPL Tasmania champions, whose four cup titles included wins in 2016 and 2018.
"At some point you are going to play a good side whether it's in the first round or the final," he said.
MORE SOCCER:
"We're probably going to go in as underdogs because we are playing one of the top sides in the competition, but it's an opportunity to show that we are not here to make up the numbers.
"We are at home so that's good."
Ken Morton's South Hobart, who have won half of the last 12 Lakoseljac Cups, face a trip to Birch Avenue where United will relish the chance to step up from the Northern Championship.
"I'm very happy with that," beamed United coach Glenn Reading.
"You always look forward to playing the top sides and they are a well-organised club who we have a good relationship with."
The two teams played a practice match last month which South won 4-2 in front of a good turnout.
"A few of our boys said they had not played before a crowd like that for a practice game so we'd hope to get another good crowd in for the cup tie."
Riverside and Northern Rangers both face journeys to the capital with Alex Gaetani's side travelling to Southern Championship side University and reigning Northern champions Rangers due to face NPL Tas side Glenorchy.
The women's Statewide Cup limited the Launceston teams' travel demands with City and Rangers drawn together and reigning Northern champions Olympic welcoming Metro to Windsor Park.
