Blue Tier Bioblitz organiser Louise Morris said participants would take part in surveys to find and identify as many species as possible in the area over a short amount of time.
Ms Morris said the Bioblitz would focus on areas of glacial refugia ecology.
"This is an area that didn't freeze over during the last Ice Age," she said.
Attendees of the event will join a team of scientists, arborists, and educators volunteering their time to study this special part of the state.
The weekend's program includes field surveys of amphibians, tree canopies, flora and fauna, aquatic life, bats, and owls; nature journaling and photography; and, a gig put on by Monique Brumby at the Weldborough Pub.
"It's going to be a really varied 48-hours," she said.
"It's for people who are really interested in the deep science, and for people who just really love the forest."
The Tree Projects' Dr Jen Sanger said the project was critical for learning about the natural values of the area.
"We can only properly manage an area if we understand what species live there," she said.
"This is especially important considering that a lot of the forests in the Blue Tier areas could potentially be opened up for logging in April.
"The Blue Tier is a particularly important area for conservation.
"The area contains internationally listed wetlands, Gondwana remnant forests containing giant trees and is home to Tasmanian devil, masked owl, spotted tailed quolls, Tasmanian wedge tailed eagle, and the endemic North East Highlands forest snail.
"There is a very good chance that we may be able to find new threatened species in the area during the Bioblitz that may not have been recorded here before. It's really exciting."
Ms Morris said the Blue Tier was a "special, special place".
"We want to save the forest with science," she said.
Free camping will be available, and attendees are asked to dress for conditions.
Places in surveys are limited so registrations are required.
