The showcase event will also be the final round of the Enduro Tour, which will play host to a stacked field of elite riders in male and female categories.
The Asia-Pacific Continental Series allows riders to gain world ranking points for the 2020 Enduro World Series with the top three riders in each category progressing.
The continental series, which runs across three races in Australia, New Zealand and Thailand, culminates with the crowning of the Asia-Pacific Enduro Champions in Thailand on November 30.
With the EWS visiting Tasmania earlier this year, Derby was once again crowned with the best trail of the year.
With around 200 competitors, Derby will be transformed into a hive of mountain bike activity.
The event will be free to spectators with plenty of activity on offer throughout the town.
Entries are still open for all levels of racing across the weekend, including the Challenge 80 which is a shortened, more relaxed, version of the main categories.
Launceston's multiple national mountain bike champion Rowena Fry again flies the home-state flag.
Having taken out the bronze medal in the elite women's category at the EWS in Derby in April, Fry is dominating the elite women's enduro field in Australia but is expecting stiff competition from Shelly Flood on the Blue Derby trails.
"Standing on the podium in my own backyard for the Enduro World Series was amazing and I am still on a high," she said.
"I am really looking forward to racing next weekend and it's great to see so many female riders racing.
"The social aspect of the sport alone has toned down the seriousness and creates a really fun weekend on and off the bike. It will be fun racing and showcasing my local Derby trails."
The North-East's EWS under-21 winner Fennella Harris will take to the trails again on Sunday with strong competition coming from Launceston's Izzy Flint who was sidelined from this year's EWS Derby round due to an injury sustained in the previous round in New Zealand.
South Australia's world cup downhill regular Connor Fearon, Enduro Tour opening round winner Jordan Prochyra and crowd favourite Paul Van Der Ploeg headline the elite field.
