The challenge asks participants to hike to Kokoda trail in Papua New Guinea to raise funds for Cancer Council Tasmania.
Cancer Council chief executive Penny Egan said last year's inaugural event raised more than $28,000.
"This was a life-changing event for many of the participants last year, who walked with local adventure trekking leader, Tom Allwright and Adventure Abroad," she said.
"One participant, Andy Moylan, is walking again to support Cancer Council Tasmania - he and his wife Rachel used our services last year and want to continue to contribute."
Each participant will raise a minimum of $2000, which then goes toward cancer prevention and support services.
The challenge will take place from October 22 to 31.
