The money will fund the construction of a new base in Launceston to meet increased demand for service.
Opposition Leader Bill Shorten said the investment would improve facilities at the base which was built 20 years ago.
Bass Labor MHR Ross Hart said the service was critical for remote communities who did not have easy access to hospital services.
The new base will include a helicopter, aeroplane and ambulance service, and a new 12-bed ward to hold patients in the event of a natural disaster, pandemic or infectious diseases outbreak.
Mr Hart said the announcement built on Labor's commitments of $30 million to address elective surgery waiting lists, $15 million for a mental health hub in Launceston, and $4.5 million to ensure Tasmanians had better access to specialists.
Mr Shorten will visit Hobart and Launceston on Saturday.
