Opener Thanuka Dabare smashed 104 off just 47 balls that also ended Longford's all-conquering stranglehold over its competition.
The reigning champions were gunning for their fourth straight premiership from a sixth season decider.
But the former Sri Lankan under-19 opener, who is thought to be under T20 consideration from national selectors, left Tigers captain Josh Adams shaking his head after the seven-wicket loss.
"It was one of the best hundreds I have ever seen in a final," he said during the post-match presentations.
Dabare looked set for a huge afternoon on Saturday chasing down 151 for victory.
He moved quickly to 25 off only 11 deliveries that had included three fours and two sixes, and Hadspen were away to 0-31 just halfway through the third over.
That tempo increased further to 71 off six overs when partner Nathan Balym (12 off 18 balls) got in the act.
Chieftains' wickets finally started to fall in a dribble, but Dabare powered away.
The 20-year-old, who was starved most of the strike in the first nine overs, brought up his 50 in 20 balls next ball.
The milestone prompted a series of boundaries as 18 came off the next four balls.
Veteran Rob Moore ended Dabare's epic knock just 13 short of the premiership win.
"This innings has been my best I have had since before in Sri Lanka," Dabare, who notched his third Premier League century and fifth overall for the season, said.
"It was a good opportunity the grand final, but it was a good team effort."
Dabare had earlier helped bring Longford back from a terrific start to lose its last nine wickets for 58 runs.
Keeper Jackson Blair had initially laid down the platform for the Tigers with a patient 39 off 78 balls.
Jason Snare set the pace with 25 off 27 balls and Dion Blair with 21 off 44 before Debare (3-14 off 7.2 overs) had a hand in the top-order scalps including Blair's run out.
Longford never recovered as Hadspen captain Liam Reynolds also turned around some early stick to claim three late scalps towards the club's first top-flight flag since the 2009-10 season.
