The Spreyton-based stayer is trying to emulate Norsqui who was successful for Broadmarsh trainer Walter McShane in 2013, after running third the previous year.
Chasing his fifth win in a row, Eastender created history 12 days ago when he became the first horse for 42 years to complete the Devonport, Launceston and Hobart cups treble.
However the Barry Campbell-trained gelding has paid the price for his consistency and is equal topweight with 58kg - 2kg more than carried by Norsqui.
The big weight, coupled with a wide barrier, has seen Eastender ease slightly in the betting from his opening quote of $5 to $6 with tab.com.au.
Bondeiger, the 2018 Launceston Cup winner, is favourite at $3.80 ahead of the Chris Waller-trained Naval Warfare who has firmed from $7 to $5.50.
Eastender was floated from Melbourne to Adelaide in the early hours of Sunday morning to avoid the heat.
HEAT WINNER: Samantha Gangell won a heat of the Australian Female Drivers Championship at Albion Park on Saturday night and finished sixth overall with 48 points.
Fellow Tasmanian Hannah Van Dongen had no luck driving mainly longshots but managed a third place on a $31 chance.
Queenslander Narissa McMullen won two of the other six heats to take out the title with 79 points.
Todd Rattray will compete in four heats of the Invitation Drivers Championship at Horsham on Monday.
TASMANIA CUP: Former local star Resurgent Spirit returned from Victoria to score an upset win in Sunday night's second heat of the $75,000 Tasmania Cup in Hobart.
Unwanted in the betting at $41, he raced one-out, one-back before finishing strongly to beat highly-fancied Victorians Sicario ($2.90) and War Dan ($3.80).
It was his 22nd win from 31 starts on the track.
