The woman was travelling north on the Tasman Highway at Runnymede near Black Charlies Opening when the car left the road and rolled, before coming to rest on its side.
The soul-occupant was cut free by Tasmania Fire Service and taken to the Royal Hobart Hospital and was admitted and treated for non-life-threatening injuries.
Traffic was disrupted for a number of hours.
Tasmanian Police said the contributing factors of the crash have not yet been determined, but were being investigated.
“Police would like to speak with a group of four motorcycle riders [that] were believed to be in the area vicinity of the crash at the time that it occurred,” they say.
“Police wish to extend their sincere thanks and appreciation to all members of the public and to witnesses to the crash who stopped to provide assistance prior to arrival of emergency services.”
