Drivers heading east toward Newstead will still be able to turn right into Lyttleton Street, past the Launceston Church Grammar school campus.
But drivers travelling west into town will not be able to turn right into Lyttleton Street, toward Racecourse Crescent.
To improve traffic flow, the council plans to add a dedicated right-turn lane at the Dowling Street and Elphin Road intersection instead.
Traffic turning into Elphin Road off Lyttleton Street would only be able to turn left, toward Newstead.
City of Launceston mayor Albert van Zetten said the changes were to prevent further crashes.
"In the five years to June 2016, there were six crashes at this intersection, two of them serious, and our desire is to see the number of future crashes in this area significantly reduced,” he said.
Feedback on the proposed changes is open until May 12 on the council website.
