The woman, 46, of the ACT, was driving a Holden Astra east on the highway near Carrick.
Her car collided with the rear wheel of a semi-trailer, travelling in the opposite direction, near the Oaks Road off-ramp, about 9.14am.
The Astra ended up on the wrong side of the road and collided head-on with a Holden Commodore, which was also headed in the opposite direction.
The female driver of the Astra died at the scene.
The driver of the Commodore, a 55-year-old man, was seriously injured and was transported to the Launceston General Hospital.
The semi-trailer driver, 57, was badly shaken but uninjured.
Authorities closed part of the highway for several hours to allow crash investigators to examine the scene.
Heavy rain added to the traffic congestion while police diverted vehicles down off-ramps.
A report will be prepared for the coroner.
Police urge anyone with information about the crash to call Northern crash investigation services at Launceston police headquarters on 63363701.
