Meander Vallery Road will be used from Prospect Vale to Deloraine and the Bass Highway from Deloraine to Devonport.
The fun run is for the Tim Blair Fun Run. Mr Blair will use the run as a way to raise awareness for childhood cancer. It is expected the road will be used from 7.30am until 6pm.
The Department of State Growth has granted the Tim Blair Fun Run permission.
