Emergency services were called to the scene about 5pm, where a ute had flipped onto its roof, a second ute and a trailer ended up on the side of the road, another ute was impacted head-on and a fourth car was hit by debris from the crash.
Police said a white Mazda Bravo ute was heading north along the highway when it was believed to have crossed onto the other side of the road and collided with the second ute, before hitting a silver Holden Rodeo head-on.
A man from the Rodeo and a man from the Bravo were taken to hospital by ambulance but were not being treated as critical.
Police said alcohol and speed were not believed to have been contributing factors, however, early investigations suggested inattention may have been the cause.
The road was closed between Woolmers Lane and Powranna Road for more than three hours.
It was the same section of highway where a crash left a Kings Meadows man dead and six people taken to hospital in December.
