The previously winless City are coming off an enthralling 3-2 extra-time victory over Southern Championship side University in Hobart.
But Ben Brookfield’s men will have their work cut out against South Hobart, who routed championship battler Launceston United 12-0.
The current top-four NPL Tasmania clubs had received favourable quarter-finals from the random draw held on Tuesday.
Hobart Zebras will return home to face state league rival Clarence United.
Rank outsiders Nelson pulled out an away fixture against Olympia Warriors, while Devonport City travels to meet former top-flight club Glenorchy Knights in a repeat of NPL and championship sides clashing.
The Lakoseljac Cup, which also acts as FFA Cup qualifiers to match-up against the likes of A-League clubs, returns over April 29-30.
