Mr Barker was shot dead outside his Campbell Town home on August 2.
His killer has since remained at large.
Last week, Tasmania Police announced it had increased the reward for information relating to his murder from $50,000 to $250,000.
On Tuesday, police announced that officers from the Serious Organised Crime Division’s cold case team and forensic services would be returning to the murder scene.
Detective Sergeant Mark Lopes said information uncovered during a recent review of the case had given police new leads.
Detective Sergeant Lopes said in particular, the team would be searching for evidence in the form of bullet casings.
“In particular, we will examine locations where the shooter may have hidden prior to and during the cowardly shooting of Shane Barker which led to him sustaining four bullet wounds,” he said.
“The evidence reviewed by Cold Case investigators indicates that Mr Barker was ambushed whilst in the process of securing his vehicle in the garage at his home on that evening.
“The evidence also supports that the murderer knew Mr Barker's movements and laid in wait for him to arrive home before commencing his cowardly crime.”
