The students are from the Fossil Free UTas group, and have said they will refuse to leave until the university agrees to divest from fossil fuels.
They entered the office at 1.30pm on Wednesday.
Fossil Free UTAS spokesman Jack Redpath said the students were equipped with sleeping mats, sleeping bags, food and water.
The sit-in follows a peaceful protest at the campus on Wednesday morning.
The university has been contacted for comment.
*The students earlier claimed to be located inside the vice chancellor's office, however the university has advised they are occupying an area in the foyer of the administration building.
